In the abstract the authors comment that "The number and levels of O-GlcNAc-containing proteins do not appear to be different between AD and control brain. The only significant change observed is a marked reduction of O-GlcNAcylated clathrin assembly protein-3 (AP-3) in AD." It is notable that significantly altered glycosylation of tau was not found. The marked reduction of AP-3 in AD implicates a defect in vesicle recycling in AD.
Post-translational modification is a characteristic event in AD and related disorders because of abnormal accumulation of cellular inclusions and extracellular deposition. There is still not much known about related enzymes and molecules other than ubiquitin and glycosylation.
