Findings
In a study that included 15,808 Alzheimer’s cases and 16,097 control subjects from multiple European and American cohorts, this allele was observed twice among the AD cases (Holstege et al., 2022).
Previously, in a European cohort of 1255 Alzheimer’s disease cases and 1938 controls, a 54-year-old Italian control subject was identified as a heterozygous carrier of the V1668M variant (Verheijen et al., 2016).
The V1668M variant is classified as likely benign by the criteria of Holstege et al. (Holstege et al., 2017).
Functional Consequences
This variant was predicted to be damaging by SIFT, benign by Mutation Taster, and probably damaging by PolyPhen-2 (Verheijen et al., 2016).
