Overview
Monoclonal antibody 1F1 reacts with mouse CLEC7A. In initial testing, the antibody was shown to work as a detection reagent in western blots.
Monoclonal antibody 1F1 recognizes murine CLEC7A. A) CLEC7A overexpressed in HEK293 cells. Western blot of lysate of HEK cells transfected with murine Clec7a, human CLEC7A, or empty vector (mock), probed with hybridoma supernatant. 1F1 recognizes murine, but not human, CLEC7A. B) Endogenous CLEC7A. Western blots of lysates of BV2 cells (left) and primary microglia (right), probed with Protein A affinity-purified 1F1. Treatment of BV2 cells with lipopolysaccharide (LPS), expected to down regulate expression of CLEC7A, eliminates the smear at ~40 kDa, but has no effect on the ~50-kDa band (*), which is considered non-specific. Generation and epitope mapping
This antibody was generated in rats immunized with a peptide from the cytoplasmic region of CLEC7A, coupled to ovalbumin. Epitope mapping has not been reported.
Specificity
On western blots, the antibody recognizes mouse, but not human, CLEC7A overexpressed in HEK293 cells. It also recognizes endogenous murine CLEC7A in BV2 cells and primary microglia.
Validation
Antibody 1F1 has been validated for Western blot applications.
Knockout validation of antibody 1F1. Western blot of brain extracts from 15-month-old APP/PS1 mice that express endogenous Clec7a (left three lanes) or mice with genetic deletion of Clec7a (right three lanes). Blot probed with Protein A-purified 1F1 (2 μg/ml). *, non-specific band. [Courtesy of Christian Haass' laboratory.]
